I've used MTT assay to assess the viability of Canada biofilms after a treatment. I had about 90% decrease in cell viability from MTT assay, but I don't get the same expressive result when I evaluate the viability through CFU/mL enumeration.
Does anyone know what is the minimum cell density MTT can detect? I found a PhD thesis that shows a lower limit of 10^3 for XTT assay on Candida. I suspect there is a limit of how many cells MTT can detect too. I could do a cell density curve and measure it, but I've got a really tight agenda at the moment and if someone else already did that, then maybe I won't have to.

It all depends on the cell line you are using. Usually you can plate between 1000 to 10000 cells per well. If the cells form clumps you can go up to 15000 cells per well.

Two things to consider here: (1) your cell line and (2) the sensitivity of the product you are using. A range I typically aim for is between 1,000-20,000 cells.
Personally, I think MTT is becoming a little outdated as there are better reagents out there to measure cell number. If you want a more sensitive experiment that looks at less that 1,000 cells, consider using a commercially available kit such as CyQUANT from ThermoFisher Scientific.
