I am trying to make insulin solution in sodium citrate buffer at pH 5.3. I notice that the solution becomes cloudy. Is this due to zinc precipitation? Will sodium acetate be a better buffer?
You can use dilute HCl (0.001-0.01N) to dissolve insulin, aliquot in portions suitable for your work and store them frozen. Insulin is quite stable under these conditions at high concentrations (1mg/ml or higher).
