At the risk of asking a stupid question. I'm planning to start isotropic fractionator experiments on insects. The protocol says to make a dissociation solution of 1L of 1% triton x-100 in 40mM sodium citrate. Unsure what measurements I need to add and how?
Not sure I can completely answer with what you have given. The 40mM sodium citrate is easy. Calculate the weight of 40 moles of (anhydrous) sodium citrate and then dissolve in water and dilute to 1 Liter total volume. Adding the triton-X-100 requires you knowing if it is a volume/volume relationship or a weight/volume (or weight/weight) relationship. Once you know that you can add 1 part triton-X-100 to 99 parts of your solution and you should be there.

I don’t disagree with John, but common practice would indicate that the Triton is a vol/vol measure. Sources for this triton-citrate reagent should indicate mol/mol or wt/wt if this were otherwise. Thus, calculate Na citrate for 1000cc at 40mM, dissolve in 960 cc (approx) HOH, add 10 cc Triton(vol/vol), bring total vol to 1000 cc with HOH.
