Hello every one,
From my last subculturing till now I'm facing with these round, spherical cells that I think are some kind of contamination. I'm using RPMI-1640 supplemented with 10% FBS and Pen/Strep. Last time I used new fresh RPMI and added “Amphotericin B” as twice as my RPMI to flask (e.g. 10µ for 5ml RPMI).
But there are still these contaminations. Would anyone help me to figure out how can I get rid of these? I circled the contaminants in attached picture.

Probably these floating cells are dead cells or unhealthy cells that cannot adhere in the medium. To figure out this problem you should decrease the concentration of your inoculate or it is better for you to change your media and if you still have floating cells then you should discard your entire flask and culture new healthy cells.

I'm afraid that these would be dead cells and I also tried with DMEM but again this contamination has happened.
Do you have any better idea to remove them?
