I have flash frozen rat brain tissue that I'm extracting protein from. Before freezing the brains were separated into hemispheres. I am looking to extract a region of interest in the back third of the hemisphere on the top. Our thought is to use a heated blade to slice the frozen tissue. My concern is that heating this tissue will result in protein damage that may skew any results. If done quickly this method should leave the region of interest still cold if not frozen. Can anyone speak to the effects of heat-altered cytokine expression affecting ELISA results?
