It is known that people use serum free medium from 2 h to overnight duration for synchronizing cells prior to experiments. My queries are as follows.

1. How serum free medium synchronize the cells? What is the mechanism? How to be sure that serum free condition has done its job?

2. Is it necessary to synchronize cells before all cell based experiments or only proliferation assays?

3. What are the other effective and less expensive agents that better synchronize the cells?

Here, cells means I am indicating cancer cells.
