I am preparing a protocol on Isolation of Candida spp from vaginal swab. At this moment, we don't have differential CHROMagar Candida medium in hand. We have blood agar, chocolate agar, general CHROMAgar. We are planning to plate the swabs directly in Blood agar.
Anyone wiith experiences on isolation of Candida spp from swab samples will be helpful. Please response.

We did extensive work on the isolation and identification of Candida albicans from a wide variety of clinical samples on Pal sunflower seed medium. We developed this medium in 1980 as selective medium for Cryptococcus neoformans. As Pal sunflower seed medium is very cheap that other media like blood agar , Sabourud agar, CHROM agar, and YPD, I advise you to employ Pal sunflower seed medium for the study of C.albicans and other yeasts in your laboratory.

You can use Sabaroud agar.For differentiation , you should use Hichrome agar .

I have worked on Candida spp before. for culturing "Blood agar" is fine almost all candida spp can grow. For differentiation you may need another media or gram stain can do the job done.
Candida spp grows well in blood agar, but you can additionally use saboraud agar and identify with gram stain.
You can use Sabouraud or YPD (yeast extract, peptone, dextrose), all Candida grow well. To see differences you can try liquid YPD + serum or 2.5 mM GlcNAc (N-Acetyl-D-Glucosamine). Another option is to grow them in YNB + Galactose and then switch to YNB + GlcNAc (fresh), at 37 degrees. C. albicans will make true hyphae, while C. glabrata or C. parapsilosis won't. CHROMAgar or other chromogenic media will help identify Candida better, but the medium is usually expensive.

I also have a similar project, isolating microorganisms from vaginal swabs, from indigenous women in the Amazon. I have kept the swabs in PBS, then in the lab spread the swabs on YPD + Ampicillin plates and also make glycerol stocks. To determine the species I am trying PCR with ITS primers and RFLP. You could do something similar.
Francisco Javier Alvarez Hi, which medium did you find was best for isolating microbes from vaginal swabs? Im using vaginal swabs for my project and I cant seem to find much info on media for vaginal cultures outside of a diagnostic investigation
There are media specific for bacteria. Candida can survive in plain water for months, so PBS is OK.
