Using nanoparticles in PTC medium has positive results in in vitro flower induction, reducing bacterial contamination, somatic embryogenesis, etc., But what're the other technical challenges encountered using different nanoparticles in plant tissue culture? and What's the possible negative impact in the edible/ crop products?
The colloidal metal nanoparticles are difficult to study because they remain partially distributed in the nutrient media. Secondly, to find a threshold value where maximal positive results are observed is critical in studying the effects of metal NPs in plant tissue culture experiments. If the concentration of NPs is raised to that threshold limit, NPs will produce toxic effects creating negative influence to the ecosystem and health of humans eating such toxic nanoproducts.
Dear Rabia Javed are you suggesting that NPs remain in the plants after they back to normal ex vitro conditions?
Challanges are how to test different concentrations of NPs for stopping bacterial infections without inhibiting plant tissue growth. If the concentration is too high the tissue will not grow because of the NPs, if it is too low, the tissue will not grow due to the (sometimes difficult to see or identify) infection. Also, you need to start your experiments with 100% infected material for statistically sound results, so you need to infect clean plant material first. How do you do that and with which source?
I think, one of the difficulties in using NPs is they form aggregates and this increases the size, than what we initially have. They could also accumulate in the plants and there is no way to see where they are localized and how much.

Using nanoparticles in medicine has triggered their application in agriculture also. I think it will have more advantage beyond the investigation but drawbacks also comes together. In PTC, treating with NPs will be taken as Np priming (especially for heavy metals) which will improve heavy metals stress tolerance. In opposite, it will have high chance of accumulation ( impactfull). So, before application it should be passing rigorous dose determination steps. I feel it like the problem with traditional medicine, both have huge potential.

I have used biosynthesized Ag and ZnO NPs in a tissue culture medium. At the optimal level of application, I got a profound response in terms of quality shoots development, reduced rate of bacterial contamination and increased chlorophyll contents, etc. But I wanted to know whether the accumulation of these NPs will have any adverse/ toxic impact on the plants developed. Metallic nanoparticles may have a positive impact on tissues in vitro but to a certain extent only. Because most of the metals are toxic to the plant cells.
