We are using the pten mouse model of prostate cancer and we'd like to determine if T-cells infiltrate the prostate and the microenvironment in the context of our experimental conditions. IHC was used to identify cd4+ T-cells. Specifically, we want to distinguish Th17 cells in the mouse prostate. We suspect we could use 2 proteins that are unique to th17 cells. I think an immunofluorescent overlay of the two proteins (1 slide) would answer our question. However, I wanted to get feedback from anyone with experience.
CD4 together with RORgammaT with overlay. If the RORgammaT does not work you could try IL-17. The problem is that other CD4 T cells in addition to Th17 cells can produce IL-17 such as Th1.

We have tried this extensively and decided it cannot be done. There are no published IL-17 stained cells in any mouse tissue that are well controlled. You must have a KO control. All antibodies to mouse IL-17A produce non-specific staining. If you get it to work, I would be very interested in learning about it.
Could you please suggest me an antibody to detect IL-17 in paraffin embedded tissues. I am not trying to stain Th17 cells. I am trying to identify the IL-17 protein in secondary lymphoid organs.
Your suggestions will be highly appreciated in this regard.
