Avoid conditions that cause cellular stress because induction of cellular stress can lead to senescence.
You should avoid the following.
A. Avoid using culture media that lacks essential nutrients, growth factors, or appropriate pH and osmolarity as these can stress cells.
B. As far as possible, avoid contamination because bacterial, fungal, or mycoplasma contamination can disrupt the culture environment and induce stress.
C. Avoid inappropriate culture conditions like low pH, high oxygen levels, temperature fluctuations in the incubator.
D. Try to complete your animal culture work in minimum possible time and return the culture vessel to the CO2 incubator.
Also, during subculture, do not expose the cells to air for long, especially during change of media and trypsinization.
Please note that senescence of cells also occurs following an extended period of proliferation in culture or in response to various physiologic stresses.
For instance, most types of primary normal cells that are grown in culture do not proliferate indefinitely. Instead, after a period of rapid proliferation, their division rate slows, ultimately ceasing altogether.
