We recently bought Dengue NS1 antigen and monoclonal antibody at IIT KANPUR.I need to know the thawing temperature and how to thaw the NS1 antigen and antibody.One of my labmate thawed at 35 degree centigrade for 15 mins at vacumn oven.And made aliquot time taken around 45 mins at room temperature and kept rest of the samples in deep freezer at -20deg C.Is the process ok?Is the proteins ok?
From my point of view, this is OK. However, each antibody and each protein have different properties and therefore it is possible that this general protocol is not optimal in your case. For example, some proteins are extremely sensitive against freezing and should not be frozen again. In this case, it is better to add 50% of bio-grade glycerol and keep the antibody or other protein at -20 degrees C. With the addition of the glycerol the solution will not form water crystals.
