: I need to test out various conc of PGRs effect of my explants. Was thinking it would be easier to add the pgr's in solution onto the top of the agar the explants are on. So for example if I wanted to bath the explant on agar that has no pgr's I would have to add 10ml of pgr in soution made from 1mg/200ml so 1ml = .0005mg or the same as if I was using agar with .5mg pgr/L. This way I can after a period of time syringe off the pgr solution on the agar and replace it with a different solution of pgr's to see the effects. Is this a reasonable approach? Positives? negatives?. Has anyone try this out? .thanks steve
hi Dhia, thanks I briefly looked at the thesis but it looks like the seedlings were started on agar and transferred to liquid medium. I want to add liquid medium to the agar ..just enough for the bottom of the explant to be in contact with the pgr's. Does this thesis address this? thx steve

thanks ricardo I'll modify this for my in-vitro jars. I'm hoping to use this method to find the right media to trigger embryogensis but rooting would be the next step

I have his first vol on pdf but the second one is real expensive will try the library again to see if I can find it...thx
