Could I use OPA derivatizing reagent and use in UV detection. I read bibliography and frequently excitation lambda is about 340 nm so I am thinking in use this lambda for UV absorption.
Yes you can use o-Phtalaldehyde (OPA) to enhance detection and we do it often when a FLD may not always be available (for client applications). The thiol functionality can still be monitored at ~ 230nm (which is usually better than the signal obtained at 340, but if you have a DAD, and you should, you can monitor and use BOTH wavelengths).
FLD detection will result in far better sensitivity, but the resulting UV signal is improved. BTW: We perform this precolumn derivatization using the automated injector's program table (very useful for sample prep type applications like this).
What does "Thiol functionality" means?. OPA reacts with amino groups only, not with OH groups.
