A colleague of mine is trying to develop a derivatization reaction to determine the amount of lenalidomide in rat plasma samples by LC coupled with fluorescence detection with the aim to develop PK/PD modeling.
He´s tried the derivatization with fluorescamine, but unstable reaction products are formed and finally, unstable chromatogram peaks make for incompatible quantitative analysis.
Does anybody know how I can help him with this step based in derivatization for FLD detection?

I would imagine that the aniline ring will fluoresce without using the fluorescent derivative. I use BSTFA (bis(trimethylsilyl)trifluoroacetamide) for derivatizing phenols for separation on GC. It may be a cheaper option to allow for separation, but that is dependent on the column you are using, and would also eliminate the possibility of using water as an eluent.
I use MSTFA+TMCS to derivatizing drugs of abuse for separation on GC-MS/MS.

Lee and Ivo, we can´t use GC with MS/MS detectpr because we don´t have them. We work actually on LC coupled with FLD os UV/vis.
since Lenalidomide contain primary and secondary amine group you can derivitize by 4-Chloro-7-nitrobenzofurazan (NBD-chloride)
Ok, but, a derivatization step is a tedious procedure with controlled experimental conditions. Do you know how to use it? Ratios, incubation, pH...
