I am pondering how to fix a decellularised organ slice to a glass and/or plastic surface with a minimal amount of substance used so it remains adequate for a perfusion set up. The issue is, that the slice will start to float upon the addition of culture medium (or any other fluid for that matter).
We are considering incubating the slice in fibrin and subsequently hope that it will attach to the surface, but we have not yet tested it. Alternatively, we could use tissue glue to adhere it to the surface manually (but this is likely to cause flow disturbance in the perfusate).
I would be very happy for advice or past experience with this issue.
If you make plate surface rough and reduce the amount of medium just up to the height of your organ tissue, media will still cover surface of tissue.
I wanted to grow cells from tissue pieces without using enzyme digestion, the tissue pieces will like to float, I scratched the bottom of plate with a pasteur pipette that helped tissue to entangled on plate cracks, I reduced the media to minimum that stopped the tissue tendency to float, cells start growing from tissue pieces, that served my purpose.
