Recently, we are conducting polarization experiments on Raw264.7 macrophages.
It is known that Raw264.7 is a classic mouse macrophage cell line that can be induced to polarize to M1/M2 phenotypes. In the literature, IFN-γ and LPS are commonly used to induce M1 type, and IL-4 and IL-13 are used to induce M2 type.
Do you have any recommended cytokine combination schemes (such as concentration ratio, addition sequence) that have been verified to be effective in Raw264.7?

IFN-γ and M-CSF can induce cell polarization, as follows:
IFN-γ:
Under the stimulation of LPS and IFN-γ, RAW264.7 cells can polarize into M1 macrophages. During this process, the cells will show dendritic characteristics, and the expression of inflammatory mediators such as iNOS, IL-1β, IL-6, TNF-α and CD86 will increase significantly (PMID: 20439986).
M-CSF:
When it acts together with RANKL, it can induce RAW264.7 cells to differentiate into osteoclasts. During this differentiation process, cells will undergo a series of changes, including actin reorganization, activation of signaling pathways such as Src and ERK, and upregulation of osteoclast-related genes such as TRAP, calcitonin receptor and cathepsin K (DOI: 10.1016/j.jff.2017.02.018).
