We have been working on breast MDA-MB-453 cells. We found that hormonal treatment increases expression of three genes: Slug, RGS2 (regulator of G protein signaling 2), and MMP-13. When cell were transfected with siRNA targeting Slug or RGS2 or two specific miRNAs (separately) that target MMP-13, the protein level of these genes increases after 3 days rather than decreases. This is true for untreated and treated cells. Does anyone know what is happening? is this a feedback mechanism specific for these cells? is it a siRNA phenomenon?
