I am looking for bovine serum that has been depleted of thyroid hormone (both T3 and T4). Does anyone know of a commercial source or a straightforward way to strip bovine serum of thyroid hormone in house?
The classical way would be e.g. stripping with Dowex resin
e.g. http://www.pnas.org/content/78/4/2230.full.pdf
But there are also some "synthetic" supplements out there, which may be able to replace FCS in your cell culture (depends on cell line) .... Some of them will include thyroid hormone as a compound, so you have to contact the manufacturer
I don't want to publicize any commercial brand but we have recently used the Charcoal and Dextran stripped FBS serum from Invitrogen (Bravo SB et al, J Clin Endocrinol Metab 2013). This serum presented ten times less FT3 and three times less FT4 than the normal FBS.
@Johannes W. Dietrich: Just an unspecific Ionexchanger...therefore I'd guess: not very specific to TH...you might loose a lot of other molecules
@Sarah: A colleague of mine is working on TH-reporter assays for a long time. Therefore “background” by THs from the FCS is a big issue for him. His overall conclusion is (as far as I remember): Medium without phenol red and switch to synthetic supplement, if applicable.
