I hope this question is not too naive, as i am new to RNA research: I am currently trying to detect putative microRNAs by Northern Blot via radiolabeling (predicted in silico by sRNA transcript data). As a validation experiment i am looking for a method to non-specifically radiolabel total RNA on membranes by an approach analog to the random priming method used for DNA.
Any suggestions, observations or general advice in that aspect would be greatly appreciated, thanks in advance !
I have seen that your question has not been answered yet. I understand that you want to get randomly labeled cellular RNA? If I understand you right you should isolate RNA and generate antisense probes (DNA) with reverse transcriptase and random hexamer oligos. However I don't get how you will find out whether your signals on the blot are specific.
May be I didn't get your real problem.
