I have measured in a cuvette my E.coli culture diluted down to OD of 0.01 and also pipetted the solution into a microtiter plate and read at OD595 (also read in spectrophotometer) but my calculations don't seem to give 0.01. Here's my method

1. 900nm reading = 0.04250; 975nm reading = 0.05520

2. Take away both numbers --> difference is 0.01270

3. Divide by k factor (used default 0.173) --> 0.073410405

4. Multiply by 10 to give path length --> 0.734104046

5. Path length minus the k factor --> 0.561104046

6. Raw data multiplied by answer from no. 5 --> 0.0427 x 0.561104046 = 0.023959143

Could someone see if there is a mistake in my calculations as this answer should be 0.01.
