Hey everyone, so I am trying to perfect my procedure for the clot retraction analysis and I am struggling to keep the clot on the meniscus line. I am treating my cuvettes with pluronic to prevent the clot from sticking on the sides and my pipetting is precise enough so I am making sure that I don't pipette way into the solution. The concentration of CaCl2 and thrombin is 2mM and 0.2 U respectively, this has been changed a lot with time. I worked with 20mM and 1 U, 10 mM and 0.5 U and now I am doing 2 and 0.2. The total volume is 500 ul (which was once decreased to 300 with 10 mM and 0.5 U Cacl2 and Thrombin respectively). Many different papers are using Triton X as a lubricating agent but I used Pluronic in my case.

It would be great if anyone could help me solve this.
