I am planning to do stem cell differentiation studies, can anybody let me know what are the suitable markers i have to look for in during germ cell differentiation in to germ layers and organogenesis and is their any cell line available for the study
It would be nice to know the types of cells you want to use. If you use ES one of the easiest things to look at is differentiation to cardiac muscle, you can see the cells on the plate contracting sincronously..
if you are using adult stem cells you will have to look for markers specific of the tissue.

I like to go with ES cells; fact i like to study the effect exactly on all the three germ line differentiation and organ development for the layers specifically Neuronal ; Cardiac and liver
for cardiac as I mentioned you can visualize them contracting. For neural you can use antibodies Tuj1, GFAP and nestin. I do not know for liver
Beside the antibodies mentioned by Dr. Garcia-Sanz, I also use Pax6 as a marker of stem cells precursors. For hepatocytes check this:

Did you still see PAX6 positive cells on the differentiated cultures?
No. I used it to detect neuronal stem cells precursors. Once that these are differentiate PAX6 goes off.
Yes Daniel, I agree, but that is the type of system where my question comes. If the cells differentiate the marker goes off, but if you differentiate the culture, do you still have distinct cells that would express PAX6, and therefore retain their progenitor status? I believe the data would be YES, but I have to data to prove it, However, some of you guys may have it.
