So you want to compare the distribution of expression between 2 groups of genes (highly expressed vs lowly expressed) and get a statistical way to determine if the observed differences are significant or not?

Then I would suggest to plot the distribution of expression of the two groups with notched box-plots and do a Wilcoxon rank-sum test to determine if the differences are significant.
