I have done ABTS assay for four fenugreek extracts (Methanol, Ethanol, Chloroform and Water). what I want to do is find out is how different extracts affects the inhibition activity? For each extract I have prepared 50, 100, 200, 300, 400, 500 microgram per mL concentrations and added ABTS solution. Do i need to do one way anova or two way anova?
I think you need to do a two way ANOVA as you have two sources of variability in your ABTS data. Your two independent variables will be Extract type and concentration of extract. The two-way ANOVA makes sense because the extracts from the different solvents are likely behaving differently (interaction effect), that could elevate the error term and mask the real effect of the different extracts or the concentrations if done in a one-way ANOVA.
It is generally recommended to go for multiple-way ANOVA and select the most meaningful data later, because the Multiple way analysis reduces your error term and magnifies the true effect of the variables. Other wise, the interaction effects would be either added to the treatment and gives you more or even false effects that leads you to erroneously claim an effect that is not naturally there and make you reject the null hypothesis (Type I Error), or the interaction term is added to the error term and masks an existing effect that leads to keeping a false null hypothesis (Type II Error).
