How long do I need to starve cardiac cell lines (e.g., AC16 and RL-14) before treatment with hypertrophic agents, specifically angiotensin?

Even before the Angiotensin II stimulus I changed DMEM to a 16-18h starvation but I guess there was no AT1/2 receptor in AC16 cells. Please, try 10 uM +/- Isoproterenol

Thank you very much for your answer! We tried starvation times ranging from 30 min to overnight and nothing worked (for angiotensin). We also tried Isoproterenol but at higher concentrations (10 mM), it was better but still not all the hypertrophic markers consistently increased. May I ask how do you assess for the hypertrophy? Do you do microscopy only or also measure hypertrophic markers gene expression?
