I'm testing the effect of a drug on cardiomyocytes. The treated cells are then used for Akt and Erk ELISA. Can anyone tell me how do I interpret the pAkt/Akt or pErk/Erk ratio? For example, the Akt ratio increases with time for a particular dose of that drug. What does that mean?

pAkt is the active form of Akt. An increase in the pAkt/Akt ratio means that the protein is active and that probably the PI3K/Akt pathway is active.
pERK/ERK is the same thing but in this case it is indicative of the MAPkinases pathway activity.
Actually, an increase in the phosphorylated fraction of a phosphoprotein indicates the activity of an upstream protein kinase. Phosphorylation of either AKT or ERK is necessary, but not sufficient for their activation. Both AKT and ERK are protein kinases themselves, and their activity can be estimated from the phosphorylation of their substrates.
