I am planning to see if N-acetyl cysteine treatment can rescue my anticancer drug mediated cell death. These are going to be 48-72hr long experiments in cell culture. I plan to pre-treat my cells with 20mM NAC for around 90 mins followed by drug addition. Under such circumstances, is it known whether or not I should add fresh NAC to my cells till 72hrs? Or does the effect of NAC stay put for long enough till 72hrs? Any suggestions or recommendations?
Yes. It should be re furbished after 24 hrs as it is an unstable compound.
hi, you are careful, because it may be toxic dose greater than 10 mM NAC. You should wash three times with PBS, after pre-administration of NAC.
NAC slowly oxidizes. In cell cultures it undergoes thiol-disulfide exchange reactions with albumin present in the incubatio buffer. You can simply measure its concentration with DTNB and evaluate its oxidation during your experiments.
