I'm looking at BCL-2 in cultured cells. I'm comparing treatments that appear to inhibit apoptosis. I'd like to look at BCL-2 levels in whole cell lysates, but I'm wondering if I should be looking at mitochondrial BCL-2?

Derek i think it depends on the purpose of the study if you are interested in studying the differential expression of Bcl-2 then it should be monitored in both fractions....however if you just want to see the induction of apoptosis after drug treatment whole cell Bcl-2 level is enough to be taken under consideration...

I agree with Thomson. It depends on your research interests. If you want to go deep into the mechanisms, you can evaluate BCL-2 levels in different parts of cells. You can simply use confocol to see whether the distribution of BCL-2 in cells are different upon treatment. Alternatively, you can separate subcellular fractions of cells, and measure BCL-2 levels respectively.
However, if you just want to see the changes of BCL-2 after treatment, I think whole cells are enough.
Thank you both for your input. Since BCL-2 isnt secreted and doesn’t undergo post translational modification, I was unsure if I needed to look at different fractions to see differences.
