You should to use PCR since in agreement with some data application of IHC in tissues could give false-positive results. Moreother, I recommend you to apply WB too.

Usually we do PCR Array first (customized plate, you can design 96 biomarkers for a plate in 96-well plate), and when we get significant changes (in our case, chemokines and their receptors), we do RT-PCR and ELISA to confirm. We also do WB and IHC for final confirmation.
