Hi everyone! I wish to investigate the alveolar macrophage (MH-S) cell line response to cigarette smoke extract. Can anyone advise if I should remove 2-ME (0.05mM) from the full media? Was wondering if the presence of 2-ME may render the cells less responsive to lower concentrations of CSE. Thanks!
clearly 2-ME protect cells against oxidative stress and increases cellular GSH content, so if you want to test the effect of cigarette smoke extract on your alveolar macrophage cell line, you probably should avoid 2-ME in your culture medium and compare the metabolism and the viability of your cells in the presence of the cigarette smoke extract with those of untreated cells grown in 2-ME free medium. Thereafter, testing your cigarette extract in the absence or presence of 2-ME would probably give you additionnal informations on the mechanism of action of your extract and the putative role of cellular defence against oxidative shocks.
First thing is 2-ME is required in the medium for some cells to combat excess ROS production by the presence of excess oxygen in vitro. So you really need to know how the cells behave if you remove 2-ME or you can simultaneously do the experiment with and without 2-ME and compare the results. Goodluck
