I'm interested in isolating neutrophils from human whole blood for subsequent RNA extraction for downstream purposes like qPCR.
Just wondering if anyone is willing to share their protocol/work flow for this method? We have previously isolated neutrophils using polymorphprep and immunomagnetic depletion, however we have not attempted RNA extraction yet. Any tips or tricks for preservation of the neutrophil cell prep so that minimal changes occur in the RNA profile? Any suggestions on anticoagulation tubes needed/blood volume/storage conditions would be very much appreciated!
