I am working on mice sertoli cells. But I have some confusion in counting age of these cells. I kill the mice, collect the sertoli cells and culture on the same day(7th day). But after culture, it takes about 2-3days or more so the plate becomes full. So usually on 9th day of the mice age I extract the RNA by kit. My question is, this RNA is of 7days or 9th days?
The cells are 9 days old even when they are not in the mouse anymore. So I would say your RNA is is from the 9th day.
Hi @Rumani Singh, Live sertoli cells usually attached with plate after 4.5hrs, but if your cells are not attaching it means that the cells are dead and floating in cultural media. Check your protocol again specially the incubation time after adding Trypsin. In my 3 years experience I never faced this problem. Need your experience, hope this will help.
