Purified preps of IgG (via Protein G column) have yielded multiple bands under reducing conditions (mercaptoethanol, 5 mins, 95c) on SDS PAGE.

Could you please help me identify the bands by looking at the attached pictures?

*please excuse the poor quality 'gel 3' - it is currently destaining.

I expect bands at 50kda (heavy chain) and 25kda (light chain).

  • I have multiple bands for each prep, which appear at the same molecular weight
  • My best guess is that I have H+H, H+L, H+L+L combinations, etc b,ut i am not sure why I am seeing a band ~65-70 KDa

I can also see a strong band ~13-15kda on one of the gels, which I anticipate is contamination.

I have altered the reducing conditions and yielded similar results.

I am struggling to find in the literature examples of SDS-PAGE where different IgG subclasses and other immunoglobulins have been denatured and compared.

  • I know that iGg3 has a long hinge but I do not know what difference in kDa to expect in this context.
  • It has also flagged that 2 bands for the heavy chain can be expected with glycosylation - if you can elaborate, i would also be grateful.
