Hello everybody. Acctually I am work with rotenone and T98g cells. Surprisingly, in a moment the rotenone does not kill my cells. I have try with other rotonone, I have change the confluence, the concentration, and simplily the rotenone does not work. Someone has hadthe same problem?
Cells in culture never really like to use their mitochodria. Cancer cells in particular can be quite happy relying on anaerobic glycolysis exclusively. You might try to estimate the % glycolysis they're using by measuring lactic acid production, or simply pH change. Not super accurate, but it could give you an idea.
