My Ph.D project is focused on a protein that is essential for viral infection in target cell types but restricts infection when highly overexpressed and in cell types with high endogenous expression. I am currently performing experiments in a CRISPR KO cell line in which endogenous expression of the gene is eliminated and am trying to perform rescue experiments. However, the gene in question is robustly expressed from a PQCXIN vector and shows a very modest rescue phenotype following plasmid titration to minimal delivery levels. Are there any retroviral vectors that are optimized for low/modest gene expression? I've looked at inducible vectors, but they all seem to be on/off switches with strong expression.
