one week ago I had revived Ramos cell line in 10 cm plate by using RPMI 1640 Ramos medium. According to protocol, Ramos media should be 4500mg/L but I used 2000mg/L glucose. Next day I changed the medium because the cell count was very low and till date cells are in the same medium. Cell count is decreasing day by day, I need confluent plate for further experiment. Kindly suggest me how to troubleshoot this problem ?

I used RPMI 1640 complete medium for Ramos with 10% FBS and 1X pen strep. incubated at 37c , 5%CO2.
The composition of medium you can find from ATCC website.

1. You added Glutamine also??
2. FBS you used was Heat Inactivated & US origin??
