Hello everybody, I'm working on ddPCR and I am needing gBlocks as positive controls of some pathogens fragments. Im going to order the gBlocks to the company IDT so I need some recommendations in order to have good fragments.
I have used gBlocks (IDT) as positive controls in dPCR. In my experience, serial dilutions are needed because the stock is highly concentrated. If you need further details, feel free to reach out.
Regarding my experience working with gBlock, they are useful for many experiments, as a positive control for example. But use these dsDNA blocks for experiments that require much more precision, for example, to optimize a standard curve, are not very useful; The reason is that being such small fragments, they are very susceptible to degradation. On the other hand, gBlocks is a cheap and fast option. It all depends on what experiment you want to use them for.
