I have a simple question on quantitation using mass spectrometry with an example.

Here is an example:

Experimental: 50mg sample was used to extract metabolites, the sample was extracted, dried, and resolvated in 150ul solvent before injecting into mass spectrometer? 

Analyte concentration = ?

Analyte signal intensity = 10,000 

Standard concentration = 100 ug/ml

Standard signal intensity = 100,000

RF = 100,000/100 = 1000

Analyte concentration = Intensity/RF = 10,000/1000 = 10ug/ml

So my question is if I dissolved had dissolved my sample in 200ul solvent prior to injection into the mass spectrometry, where should I use 200ul. My concern is the resolvation can be done at different volume- 200ul or 500ul or other. How does this matter in quantitation? Where should I use this volume in quantitation?
