I have infected some mice virus, harvested their blood and aim to quantitate virus titer using real time pcr. May I know whole blood or serum sample is more suitable for this case?
im looking at enterovirus, which is one of the picornavirus. So that means we have to decide base on whether the virus may attach to some bloods cells not is it?
In general, blood virus levels are mainly represented from the plasma. Whole blood is not a good idea since the cells may contain a lot more copies of viral DNA from immature virus which can interfere with your actual virus titer. And also, serum is not a good option as the clotting process entrap some of the free virus and lead to a reduced virus compared to the actual virus titer. I would suggest you to use EDTA to prevent clotting and collect the plasma for your analysis. I hope this helps. Good luck.
