I have used Image J, Photoshop and Vision Works software to compare protein band densities in x-ray images from ECL-WB, yet there was a huge discrepancy among the results.

I think that variability of results are attributed to the tools of selection of the band area which is subjective and hard to reproduce. Therefore, I would like to know whether there is a more objective alternative method to measure subtle differences in band density. A method to read the band density from either an X-ray film or PVDF membrane in a more objective / automated manner taking in consideration both band size and color intensity pixels with subtraction of the background noise.
