If I do a dialysis with the help of a dialysis membrane of my protein and then perform SDS-PAGE to check my desired band...... Excise it and purify... Can I do the sequencing of it??? As I am unware of it... Will be grateful with the guidance.... and the procedure to use it.

If it is just for N-terminal sequencing, I'd simply blot the protein onto PVDF (not nitrocellulose!). Stain with a removable dye (usually Ponceau S), cut out the band of interest and use in the sequencer directly.
