I'm curious to know if anyone has a cell culture protocol for a co-culture of AML primary samples with either stromal or endothelial cells? I have very limited samples so no room for "trial and error". I'm assuming they can be thawed as you would any frozen cell vial and resuspended in PBS, but what media has worked? With our cell lines in co-culture, we use EGM + 10% FBS, but I don't know if this will support the growth of primary cells? I am also analyzing the population that adheres to the stromal/endothelial layer but don't know if trypsin will be too harsh? Are these cells more delicate than cell lines in terms of centrifugation? Any information of what has worked would be greatly appreciated.
