I want to prepare bacterial samples to observe the bacterial cell lysis by bacteriophage protein (endolysin) through scanning electron microscopy. Please, anyone can share the protocol of bacterial sample preparation for scanning electron microscopy. Thank you so much.

If your sample is in suspension: Apply a small volume of concentrate on to a filter of 0.22 or 0.4 micron filter, Merck Millipore Millicell Hanging Cell Culture Insert or similar one. Allow time for absorption on to the filter and the rinse 3x with cacodylate buffer followed by fixation in 2% glutaraldehyde. Rinse off the fix in cacodylate buffer. Follow any standard protocol for dehydration in ethanol and dry in HMDS or CPD. Mount the membrane filter on to stub using carbon tape and sputter coat. Good luck!
