I have isolated protein from PBMC by using RIPA buffer. I want to precipitate my proteins by using Chloroform/methanol precipitation method. I want to know whether precipitated proteins by using this method interfere in ELISA?
Hi Gyanendra, I think there are 2 factors to be considered using chloroform/MeOH precipitation of proteins for ELISA. 1) the organic solvents will probably denature the proteins and aggregate them. Proteins can be resolubilized but any conformationally specific epitopes can be lost. 2) removing the chloroform/methanol from the sample before ELISA. A speedvac of the samples should do the trick.
i do not recommend this approach as you will introduce lots of problems. Rather try to use an ELISA protocol that has sufficient sensitivity for directly detecting your Analytes. If the ionic detergents (SDS and sodium deoxycholate) still give you problems in the ELISA look for other extraction buffers and protocols, or try if you can work with diluting your sample.
