Wondering if anyone has any insight on why my gels are not running well? They are bowing in the center when running and it constantly switches itself off (the power cell will say that the load is not detected but it had been running just fine for 10-15 min).
Notes: It is the bio-rad mini protean system. I have checked all of the parts and when I run checks with precast gels they run perfectly (they are older gels and the percentage is not correct so I do not use them). The only problem I have is when I run my own handcast gels. I have been making gels for years with no issues and I have the same recipes that have worked well for me. When I run my own gels with the samples I have (similar samples run just fine on my other equipment), the gel bands bow and the powercell constantly throws an error.
Buffers are brand new and made fresh every time. All equipment is cleaned between uses. I have worked hard to see if there are any leaks with my gels. I run at 100V until the bands have run to the bottom. The samples are not high in any salts or urea. Gels do not have any bubbles.
Any ideas why my gels (that have been fine everywhere else) are not working here? Any tips? Thanks so much!
