I am facing a problem to generate a stable cell line after successful transfection. I added puromycin to kill the untransfected cells, however, eventually all my transfected cells are also get killed. I am not sure why my transfected cells could not survive. Apart from lowering the antibioctics concentration, do you have any suggestion for me to troubleshoot this problem?

All of the cells may die due to low transfection efficiency. How many cells do you get transfected?
