Hi, I'm working on C2C12 myotube differentiation using DMEM (Horse Serum 2% and PEST 1%) and I've noticed that the cell culture looks really dirty.
Here's my picture of c2c12 on day 3 of differentiation and i don't quite get why i'm seeing all these debris (they actually look like dead cells to me).
SO I tried washing cell culture with PBS 1X, but these debris just won't get off. I would really appreciate if someone could tell me what the problem is and how to solve this problem. Thanks!

I'm going through the same problem, and my C2C12 is looking the same as yours. I've tested other glucose concentrations in the culture medium, horse serum aliquots, cell freezes and nothing!
The only thing I've noticed that can explain is the presence of sodium pyruvate in the culture medium. When I was successful in differentiation previously it was in a medium without sodium pyruvate, and now that I thought that might be an explanation.
Were you able to figure out what caused your differentiation?
