Problem regarding microfluidic culture of MCF 7 spheroids?

Dear all,

I am attaching pictures of microfluidic culture of MCF7 on day 2, day 3 and day 5. I found on day 2 the spheroids had tight boundary but the next day all of a sudden the spheroids are loosing their integrity. I kept the chip undisturbed between day 2 and day 3. On the day 5 I found the microwells had brownish colour and spheroids were not visible in microscope. Please guide me if the spheroids are loosing integrity due to acidic pH or some contamination issue or some other issue?
