I have recently started cell seeding inside my PDMS microfluidic device with syringe pump. I am actually trying to form tumor spheroids inside microwells coated with pluronics. However, the MCF7 cells instead of forming spheroids are attaching to PDMS surface. Please let me know if you have faced similar problem while forming spheroids on PDMS microfluidic devices

Cell spheroids on-chip or anywhere else needs to the anti-adhesive substrate which cells are not able to attach to them, any way you are going to use pluronic as I used it before and could not solve my problem to spheroid generation on PDMS well, I think you should covalent immobilized pluronic on PDMS by changing surface chemistry, or play with concentration, or change your anti-adhesive agent (best way), good luck

Hadi Hashemzadeh I tried incorporating pluronic in PDMS and then made the chip from one of the protocols. However, still there is some cell attachment. Can you tell me how did you solve this problem in PDMS devices. Thanks in advance!

try to step by step, and layer by layer of your materials (Pluronic) and dry in oven, then inject next layer then dry it, repeat several times to achieve best result
