I have a problem with the solubility of the synthetic cannabinoids with CUMYL-based structure in DMEM medium. I’ve tried dilution from stock prepared in DMSO, EtOH or acetone, so as to obtain 200µM or less of my substances. Each of the solvents were adjusted to different final concentration 0.1-2% (v/v). All attempts failed, regardless of whether the medium was with or w/o FBS. Crystal formation I have noticed under microscope.
Any suggestion how to solve these complications?

Working with cannabinoids can be challenging since they tend to be very lipophilic. You can try adding a few drops of Tween 20 to your DMSO stock solution. I’ve also had success using a high quality grade of N-propyl acetate as the primary solvent for stocks. I’ve not used this solvent in cell culture, so best to check for toxicity first.. Regardless of the solvent used, it should not be added directly to your culture medium. Instead, add the culture medium drop wise into your solvent solution with vigorous stirring, (but not to the point of foaming) making sure all hints of precipitation are gone between additions. As the solution becomes more dilute, you can add the culture medium at a faster rate to bring your drug concentration to the desired point. Serial dilutions can usually follow without incidence. Keeping the medium warm while diluting can also help. Hopes this works for you.
Beverley A Moore Beverley, thank you for your answer. I will try to check your solutions soon. I'm also thinking about dissolving these compounds in HSA solution - stock solution and then make the appropriate concentrations in the cell medium.
